Antibodies to chlorophyll and their reactions with chloroplast preparations.

چکیده
Antibodies to chlorophyll are specifically adsorbed onto the membrane surface of thylakoids. The antibodies inhibit photosynthetic electron flow from water to NADP®. This inhibition is presumably caused by adsorption of the antibodies onto the centre chlorophyll of light reaction II. Fragments of the thylakoid membrane, obtained by ultrasonication and subsequent fractioning centrifugation, exhibit only photosystem-I activity. Conversely, the specific adsorption of antibodies to sensitizer chlorophyll has no inhibitory effect on electron transport. The ferricyanide H i l l reaction of chloroplast preparations is inhibited by chlorophyll antibodies. From these observations it is concluded that the centre chlorophyll of light reaction II and at least part of the sensitizer chlorophyll is located on the surface of the thylakoids. As agglutination is sterically inhibited by the membrane protein, it is assumed that the chlorophyll is located in gaps or pores of the protein layer. Two fractions of the lamellar system exhibit photosystem I activity of different characteristic electron donor specificity. These fractions can be further distinguished in terms of their circular dichroism and protein composition.

A reaction center preparation lacking Subunit III was obtained by means of Triton X-100 treatment and DEAEcellulose chromatography or su...
متن کاملReactions of Antisera to Lutein and Plastoquinone with Chloroplast Preparations and their Effects on Photosynthetic Electron Transport
An antiserum to lutein inhibits photosynthetic electron transport between water and potas sium ferricyanide in chloroplasts from green Nicotiana tabacum var. John William’s Breadleaf. However, electron transport between diphenylcarbazide and potassium ferricyanide is not impaired. From this it is concluded that the photochemically active carotenoid should feed its electrons into the photosynth...
متن کاملLocalization and Quantitative Determination of Ferredoxin-NADP Oxidoreductase, a Thylakoid-Bound Enzyme in the Cyanobacterium Anabaena sp. Strain 7119.
Thylakoid membrane preparations obtained from mechanically disrupted (sonicated) cells of the cyanobacterium Anabaena sp. strain 7119 show a membrane-bound ferredoxin-NADP(+) oxidoreductase (EC 1.18.1.2) as determined either by specific antibodies or by using the ferredoxin-dependent NADPH-cytochrome c reductase activity, which is a specific test for this enzyme. However, in contrast with highe...
متن کاملThe oxidation of manganese by illuminated chloroplast preparations.
It has long been known that manganese is an essential micronutrient of plants. Although the precise functions of manganese are unknown there is evidence that it may be concerned in chlorophyll synthesis, photosynthesis, respiration and nitrate assimilation. Decreased photosynthesis under conditions of manganese deficiency was originally attributed to the diminished chlorophyll content, but more...
متن کاملChloroplast Proteins
After solubilization of stroma-freed chloroplasts with deoxycholate, the lipids and the detergent used are separated from the proteins by gel filtration. In this way not denatured pigment-containing protein preparations were obtained. The particles in fraction 1 exhibited a molecular weight of 600 000 and contained an average of 25 chlorophyll molecules. The circular dichroism spectrum showed e...
